In vitro transcription assays and photoaffinity labelling of the transcriptional complex.

Methods

In vitro transcription reactions were carried out using RNA polymerase purified from lysed pea chloroplasts. The enzyme was incubated with the template DNA (5A/30T) containing the 16S rRNA promoter and a-32P-ATP (30 μCi, sp. act. 3000 Ci/mmole) and the photoprobe 4-thio UTP (200 μM) in the presence of 50mM Tris-HCl, pH 8.0, 10mM DTT, 1 μg BSA and 5mM MgCl2 at 25°C for 45 min.

Biological Activity

4-Thiouridine-5'-Triphosphate, also known as 4-Thio-UTP, is a potent agonist for P2Y2 and P2Y4 receptors. It is used in the study of crosslinking experiments, labeling of transcriptional complex. It also affects elongation and termination activity, causing transcriptional pause.
