4A3-SC8 is an ionizable cationic lipid (pKa = 6.66) and a dendrimer that is widely used in the generation of lipid nanoparticles (LNPs) for the delivery of siRNA, mRNA, single-guide RNA (sgRNA), single-stranded DNA (ssDNA), and PEGylated BODIPY dyes. LNPs containing 4A3-SC8 can encapsulate siRNA targeting Factor VII, selectively accumulate in the liver, and effectively reduce hepatic Factor VII activity in mice. Additionally, intravenous administration of these LNPs encapsulating an mRNA reporter and PEGylated BODIPY dye has been successfully applied in tumor imaging studies in mice.
