Cyanine 3-dCTP
Cy3-dCTP can be used for direct enzymatic labeling of DNA/cDNA by PCR with Taq polymerase, Nick Translation with DNAse I/ DNA Polymerase I, cDNA labeling and 3’-end labeling. Cy3-dCTP can replace dCTP in reactions in which it serves as a substrate for T4 and Taq DNA polymerases, E. coli DNA polymerase (holoenzyme and Klenow fragment), reverse transcriptase (from AMV and M-MuLV) and terminal transferase. The synthesized dye-labeled DNA/cDNA probes can be used for the identification of specific sequences by in-situ hybridization, microarray or blotting techniques. Cy3-dCTP can also be used for multicolor fluorescence labeling. Optimal substrate properties and thus labeling efficiency is ensured by an optimized linker attached to the C5 position of uridine. Recommended Cy3-dCTP/dCTP ratio for PCR and Nick Translation: 30-50% Cy3-dCTP / 50% dCTP.
