Folinic acid, clinically known as leucovorin, is a ubiquitous member of biological folate pools, usually accounting for 10 ~ 20% of total cellular folates. Folinic acid can reduce methotrexate (MTX)-induced cytogenetic damage, and appears to be an anticlastogenic agent. Folinic acid serves as a rescue agent used alone or in combination with 5-fluorouracil to decrease MTX-induced toxicity.

Applications
MTX alone induced a concentration-related increase in the percentages of micronucleated binucleated cells (MNBN) and aberrant cells (Abs). The addition of folinic acid at 50 μg/ml significantly reduced MNBN by 40 ~ 68% and Abs by 36 ~ 77%. Inhibition was also seen at 5 μg/ml folinic acid (12 to 54% for MNBN and 20 to 61% for Abs).

Chronic administration of MTX induced suppression of skeletal growth in mice. Folinic acid treatment following MTX administration appeared to reverse this growth inhibition.
